Research Abstract |
IL-1 has a variety of functions not only in the immune response but also in neuroendocrine system, affecting proliferation, differentiation and inducing inflammatory mediators. Although IL-1 receptor and its antagonist cDNAs have been cloned, IL-1-mediated signal transduction system has not yet been clarified. In these regards, we have done following studies : (1) Role of IL-1 and its action have been investigated in the human T cell leukemia cell line, HSB. 2 cells, particularly for the synergistic action of IL-1 in the induction of IL-2 and IFN-r mRNA production (Microbiol. Immunol. 1989 ; Immunol. 1990). (2) We have demonstrated that human astrocytoma cell line, U373MG, responds to IL-1 with high level production of IL-6. The action of IL-1 was not mediated by cAMP, but was augmented by cAMP, showing modulatory role of cAMP in this system (B. B. R. C. 1990). (3) In addition, U373 cells produce high levels of IL-8 and MCAF (Mo chemotactic and activating factor) mRNA expression in response to IL-1. Transcriptional regulation of these genes by IL-1 and stabilization have been studied in this system as well as several hepatoma cell lines (manuscript submitted). (4) In relation to the cytokine network, role and action of other cytokines (IL-2, 4 and 5), particularly in the context of B cell activation and parasites-induced eosinophila have been studied (J. Immunol. 1990 ; Exp. Hematol. 1990 ; Life Science Adv. in press). We believe that these studies contribute very much to the understanding of role and action of cytokines in vitro and in vivo, and to the manipulation of inflammatory diseases including infection, autoimmune and neoplastic diseases.
