Research Abstract

造腫瘍性を有するV79細胞の培養上清中には,低分子と比較的高分子の増殖制御因子があるが、この中の低分子の物質の一つは分子量2、000の熱に不安定、トリプシンで活性増強されるオリゴペプチド前駆体で、正常・悪性細胞両者に共通した増殖制御プロセスに働く制御因子であることが判明した(Cancer Res 48,3737,1988)。もう一つの低分子のものは、物理化学的にチミジンでることを証明し、チミジンは密度阻止の状態で濃度が立ち上がり部分的に増殖抑制に係わっていることをチミジンキナ-ゼ欠損株を用いた実験で証明した(BBA論文提出中)。一方、分子量約13万の制御因子は、TGFーβや腫瘍壊死因子(TNF)、ILー1αとも物理化学的にも生物学的にも全く異なり、しかもヒト線維芽細胞にはほとんど効果がないが、種々のひと癌細胞に対して抑制的に働くため腫瘍増殖抑制因子(Tumor Grovth Inhibiting Factor,TGIF)と命名した(J.Cell Science,論文提出中)。このTGIFの部分的精製の段階での性状は、熱、酸、トリプシンに安定なシングルチェインのポリペプチドであり、上皮系、間質系を問わず種非特異的で、ヒト癌細胞にも効果を有し、血清濃度に依存してBifunctionalに働き、その作用機序に関して、EGF、PDGFとの拮抗あるいは競合は認められなかった。これらの結果、この物質は腫瘍増殖に関連した全く新しいタイプのサイトカインであることを再確認した。現在,無血清による大量培養によりこの物質を大量に精製しアミノ酸分析による一次構析を行っている.
