Research Abstract |
Several clinical studies have indicated that female alcoholics may be more susceptible to the development of alcoholic liver cirrhosis than their male counterparts. We studied the effect of acute and chronic ethanol administration on the liver regeneration and ornithine decarboxylase (ODC) in male and female rat livers. 1. Male and female Sprague-Dawley rats had a 68% hepatectomy accompanied by oral administration of 5 g/kg of 20% ethanol as ethanol group. Control groups were given same calories of glucose solution. Serially at 0, 4, 8, 12, 24 hours after hepatectomy, ODC activity, immunoreactive ODC (IR-ODC) protein, DNA synthesis, liver remnant weight and total protein contents of liver were analyzed. Liver weight, total protein contents and DNA synthesis were more significantly decreased in the female ethanol group than in the male ethanol group. ODC activity and IR-ODC protein were also more significantly decreased in the female ethanol group at 4 hours after hepatectomy than in the male ethanol group. These data suggest that the acute ethanol administration suppress the liver regeneration more prominently in the female rat than in the male. 2. Sprague-Dawley male female rats were pair-fed with a high-fat, low protein diet plus ethanol or isocaloric sucrose for 6 weeks. IR-ODC protein and alcohol dehydrogenase (ADH) activity were higher in the female control than in the male control. IR-ODC protein and ADH activity decreased in the female ethanol group compared to the female control, while these enzymes increased in the male ethanol compared to male control. The highest accumulation of hepatic collagen-bound hydroxyproline was found in the female ethanol group. In conclusion, a marked decrease in the liver regeneration in the female rats may be related to the clinico-epidemiological reports that female alcoholics are more likely to develop alcoholic liver cirrhosis.
