Research Abstract |
1. Screening tests for antisperm antibodies were performed on 684 infertile women by Sperm Agglutination test (SAT), sperm immobilization test (SIT), enzyme-linked immunosorbent assay (ELISA), immunobeads test and mixed antiglobulin reaction (MAR). Seventy-one patients with circulating antisperm antibodies were selected by these methods. 2. The monoclonal antibodies against human sperm were obtained by using Balb/c mouse spleen cells immunized with capacitated human sperm. All antibodies showed inhibitory effects on Zona free hamster egg-human sperm penetration test (ZSPT). 3. Monoclonal antibody, MH61, which reacts specifically with capacitated human sperm was conjugated with Dynabeads M-450. A remarkable agglutination of MH61 coated beads was observed after incubation with sperm of fertile men, while poor agglutination activity was observed in sperm of infertile patients.
