The biochemical and pharmacological characterization on the serous and mucous cells in the three major salivary glands of rat were examined by comparison of the neutral and acid glycoprotein (GPs) species in the secretory segments collected by microdissection and saliva elicited by administration of a various sialagogue. Results : i. Electrophoretic profiles of GPs in the secretory segments : (1) In the submandibular gland (SMG). the acinar (AC) segments contained major GP, 13OK Da (SMG mucin), whereas the segmen t of g ranul ar tubules (GT) con ta ine d ma j or GP. 31KDa (Kallikrein). These GPs were stained by PAS but not stained by alcian blue (A. B.). (2) The AC segment in the parotid gland (PAG) contained GP of 94, 51, 43 and 31KDa stained only by PAS. (3) The AC segments in the sublingual gland (SLG) contained sublingual mucin stained by PAS and A. B. and 6-7 species-of GP stained only by PAS. 2. Elect rophoretic profiles of GPs in the saliva : (1) In the SMG, GPs in AC was observed in isoproterenol (IPR), dobutamine (DOB), methoxamine (MTX), phenylephrine (PHE) and substance P-evoked saliva, whereas those in GT are mainly observed in MTX and PHE-evoked saliva. (2) GPs in AC of PAG was observed in IPR, DOB, MTX, PHE and SP-evoked saliva. (3) GPs in AC of SLG were observed in pilocarpine, carbachol, bethanechol and SP-evoked saliva. These results indicate that three major salivary glands contains species of the neutral and acid GP characteristic in the secretory cells and that subtype of receptors mediating secretion of GPS are M_3, beta_1, NK_1 for seromucous AC cells of SMG, alpha_1 for serous GT cells of SMG, M_3, alpha_1, beta_1, NK_1 for serous AC cells of PAG and M_3, NK_1 for serous or mucous cells of SLG.
