Research Abstract

1.タバコ葉肉細胞が脱分化を起こす時に特異的に発現する遺伝子を単離することに成功した。この遺伝子は、G0期にある葉肉細胞のプロトプラストに脱分化を促す植物ホルモンであるオーキシンを与えると、10分で発現が検出され、4時間以降24時間まで強い発現が見られるが、細胞がS期に入る前に停止するという一過性の発現様式を示す。この遺伝子の構造解析から、大腸菌の飢餓条件で発現する遺伝子と相同性を持つことが判明し、RNAポリメラーゼと相互作用を持つことにより、調節遺伝子である可能性が示唆された。この遺伝子をタバコに導入してトランスジェニック植物を作製し、植物個体内で恒常的に過剰発現されたところ、オーキシンによって保たれる頂芽優勢が破れ、脇芽の発育が旺盛になった。これらの観察は、この遺伝子の産物は、脱分化のみではなく、より一般的にオーキシン作用の情報伝達機構の一部に関与していることを示唆している。
2.Agrobacterium rhizogenesのRiプラスミドには、植物体で発現するrolA〜Dとよばれる遺伝子が存在する。この遺伝子を植物に導入すると植物体が矮化すること、また、維管束細胞に特異的に発現すること、などがすでに本重点領域研究の研究によって明らかにされていた。一方、タバコ属の中の一群の種の染色体には、偽遺伝子と考えられていたrolBおよびCと相同的な配列(Ngrol)が存在することが知られていた。本年度の研究によって、この配列を持つ種(N.glauca)と持たない種(N.langsdorfii)の異種かけあわせによって生じる遺伝的腫瘍形成の過程を調べたところ、これらの配列が遺伝子として発現していることが発見された。この結果は、タバコ属の種の進化の過程でこれらの遺伝子が植物に移行し、細胞の脱分化、および器官形成に働いてきた可能性を強く示唆するものである。
