Research Abstract

本共同研究の大きな目的である真核生物の遺伝的組換えの分子機構の解析の成果が二つ,この研究分野では最も評価の高いCell誌に我々の研究室とKlecknerの研究室から二報掲載された(Cell69,475-470,1992)。その一つは有糸分裂期のDNA傷害修復と組換え,そして減数分裂期組換えに中心的役割を果たすRAD51遺伝子の単離とその変異株の遺伝的性質の解析結果,Rad51蛋白質に生化学的性質の解析結果,RAD51蛋白質が構造的にも機能的にも大腸菌RecA蛋白質に良く似た性質を持つこと等を報告した。他方,Klecknerの研究室では,酵母の減数分裂期組換えに特異的な遺伝子DMC1を単離し,その性質がRAD51と非常に良く似ていることを示した。しかし両者の遺伝子の機能は構造的,機能的に似た性質を持っているにもかかわらずお互いに手々減数分裂期の機能の欠損を相補することは出来ない。両遺伝子を欠失した変異株の性質の解析はRAD51遺伝子がほとんどの表現形で上位に位置することを示した。興味深いことに,一度減数分裂期に入ったrad51欠失変株の有糸分裂期の欠損はDMC1により相補されることが分かった。
本年は更に,(1)Rad51蛋白質の研究が一段と進歩し,RAD51蛋白質がssDNA依存ATPase活性を持っている事が分かり,その反応条件(ssDNAの長さ,Mg濃度MPH)がRecA蛋白質のssDNA依存ATPase活性と同じであること,(2)Rad51-ATP-dsDNA複合体を電子顕微鏡下で観察解析したところ,複合体は右巻ラセン構造を持つフィラメントを形成し,B-型DNAを1.5倍引き伸ばし,二重ラセンを巻戻す事,また3塩基対に1個のRad51蛋白質が結合し,RecA-ATP-dsDNA複合体と全く同じ性質を持つことを示した(Science259inPress.1993)。
Rad51蛋白質とRecA蛋白質の構造の違いはRad51蛋白質のN末端がReca蛋白質より120アミノ酸残基長く,1方,C末端はRecA蛋白質の方が90アミノ酸残基長い。従て,上記で述べた同じ性質を示す複合体形成にかかわる両蛋白質の領域はRecA蛋白質のアミノ酸30番目から260番目に相当する領域である。興味深いことに,この共通領域は大腸菌RecA緑膿菌RecAの間で作ったキメラーの解析で得られた中央のドメインと一致している(J.Mol.Biol.226,651-660,1992)。電子顕微鏡観察ではRad51複合体ではN末端に相当する部分が観察できなかったので,Rad51蛋白質のN末端はflexibleな構造をとっていると考えられる。RecA蛋白質のC末端部分はRecA蛋白質のDNAとの結合の強さを制御する領域やRecA蛋白質にフィラメント同士の相互作用に関連した領域と考えられている。
真核生物のRad51蛋白質がrecA蛋白質とその性質を共有するというこの知見は,Rad51蛋白質が真核生物共通の組換え蛋白質である可能性を提示した。我々はRAD51相同遺伝子をヒト,マウス,ニワトリからクーロンした。これら3つの蛋白質は全く同じ大きさ(339アミノ酸)で,99-97%同じアミノ酸から構成されていた。酵母のRad51蛋白質とも高い相同性(83%)を示した。最も興味深い事はRecAの30番目から240番目に相当する領域が共通に保存され,この領域が組換え蛋白質の基本的な,共通な機能に関与していることを示した点である。真核生物には更に,このN末端側に端側に真核生物の組換え蛋白質に共通な領域が存在していることも解かった。マウスRAD51遺伝子の発現量を種々な臓器について調べてみると,遺伝的組換えが最も活発な卵巣,睾丸,また抗体産生の活発な脾臓と胸腺,増殖な盛んな細胞胚芽細胞で最も高く,酵母のRAD51遺伝子の発現が減数分裂期やG1/S期の細胞,またX線や薬剤処理した細胞で誘導される事と良く一致していた(Nature Genetics submitted,1993)。
本共同研究は減数分裂期組換え機構の解析で始まったが,成果は遥に大きく真核細胞の共通の組換え蛋白質の同定とその性質の解明にまで及んだ。この発見は遺伝的組換えの基本機構の解析はもとより,現在注目されている遺伝子発見は遺伝子的組換えの基本機構の解析はもとより,現在注目されている遺伝子ターゲッティング技術の開発,遺伝病の治療,染色体工学技術へと大きな広がりを期待できるものである。
