細胞増殖におけるプロテインキナ-ゼC群の機能の分子生物学的解析

Research Abstract

昨年度までに確立したcPKCα,βI,βII,γおよびnPKCδ,ε,ηに加え新規θについてもcDNA発現系を確立した。また、各分子種を識別する特異抗体を作成し、種々の組織、細胞での発現を確認した。これらを用いて、各分子種がシグナル伝達系において実際にシグナル伝達素子として機能するか否かを調べるために、ラット繊維芽細胞3Y1にcDNAを導入し、TPA応答性シス因子を介する転写活性化能を指標として各分子種の機能を検定した。その結果上記4種のcPKCに加えて4種のnPKC分子種についても転写活性化能を有すること、すなわち全でがシグナル伝達系において実際にシグナル伝達素子として機能する事が確認された。さらにこの系を用いる事により、種々の活性化因子に対する各分子種の細胞内での活性化の程度を特異的に検出することがはじめて可能となった。種々の活性化因子に対する各分子種の応答性を検討したところ、cPKC群とnPKC群とで大きな差のあることが明かとなった。すなわちTPAは全ての分子種に対して大きな活性化能をもつのに対して、生理的な血清は、予想に反してnPKC群の諸分子を選択的に活性化することが明かとなった。現在血清中の因子の同定を進めている。
一方、nPKCδ,εについてはその精製に始めて成功し、これを用いてそのキナ-ゼ活性の活性化機構を検討した。その結果δがεと同様、いわゆるCaー非依存性のnPKC活性を示すこと、しかしεとは異った基質特異性、活性化因子依存性を示すことを明かにした。また種々のPKC阻害剤がcPKCと同様nPKCδ,εにも作用することを示した。
