Research Abstract |
1) Phylogenetic studies on major Japanese conifer species by means of restriction fragment length polymorphism (RFLP) analysis Chloroplast DNA of major 10 conifer species of 5 genera were analysed on their RFLP digesting with restriction enzymes of Pst I and Sa 1 I. The species analysed were Cryptomeria. japnoica, Iarix kaemp feri, Pinus densiflora, P. thunbergii, P. koraiensis, P. strobus, P. parviflora var. pentaphylla, Picea jezoensis and Picea jezoensis var. hondoensis. For pines two additional enzymes of Sac I and Xho I were applied. Phylogenetic deodrogran was made using a computer program of PHYLIP. Pine species were classified into two groups, namely hard and soft pines. RFLP relationship of other species including the pines were in good fit to those of the ordinal classification. 2) Physical map of chloroplast DNA in sugi, Cryptomeria japonica Sugi chloroplast DNA was digested with 4 restriction enzymes (Pst I Sal I, Sac I and Xho I) and estimated its genom size as 133 kbp. Total number of 36 probes and genes extracted from sugi, wheat and tobacco were used for RFLP analyses. They were as followings : 9 probes of Sal I RFLP library of sugi, 8 probes of wheat cDNA, 2 probes of tobacco cDNA, 17 probes involving 20 genes related to photosynthesis in tobacco. Sugi chloroplast DNA had no inverted repeat, and if we assume that the chloroplast DNA of tobacco was a founder, 6 events (one deletion and 5 inversion) were thought to needed to result sugi chloroplast DNA.
