Research Abstract

We have been studying on the development of osteoinductive new hybrid materials. In this time we produced a hybrid material compounded with fibroblasts which may differentiate into osteoblast and or cementoblast and porous hydroxyapatite. Periodontal ligament cells obtained from incisor roots, were cultured at 37^。C in alpha-MEM plus 10% fetal calf serum. The experimental animals were Japanese White Rabbits. Artificial separate two bone defects adjacent to different molar root surfaces were prepared in the mandible of rabbit. The new hybrid material granules were placed in the one bone defect and the other one was filled with porous hydroxyapatite granules as control. Three and 6 weeks after the operation, the animals were sacrificed, fixed in 10% formalin, demineralized in 5% trichloroacetic acid and embedded in paraffin in the usual manner. Then the serial sections 6 mum in thick were obtained, stained with H. E. and observed histologically using light microscopy. The findings at 3 weeks after the operation showed that a characteristic differenece could not be observed between experimental and control areas. On both places, phagocytes like a polynuclear giant cell attached to the granulor surfaces, and new bone began to form from the adjacent mandibular bone. The granules were partly surrounded with new bone. The findings at 6 weeks after the operation showed that some osteoid like a calcified material was recognized on the surface of new hybrid materials. On these areas the cells invaded perpendicularly into the granules. On the other hand, on the control areas these findings were not observed, and the granules were surrounded with fibrous connective tissues. These calcified materials formed on the granule surfaces may be considered to come from the periodontal ligament cells. But it would be clarified in further studies if the new calcified materials are induced by the periodontal ligament cells.
