Research Abstract

Two oligopeptides corresponding to the residue 238-252 of apolipoprotein E3(Ag 3 : GluVal-Arg-Ala-Lys-Leu-Glu-Glu-Gln-Ala-Gln-Gln-Ile-Arg-Leu)and two substituted residues 238252(Ag 7 : Glu-Val-Arg-Ala-Lys-Leu-Lys-Lys-Gln-Ala-Gln-Gln-Ile-Arg-Leu)were synthesized. These oligopeptides of Ag 3 and Ag 7 were coupled with bovine serum albumin and injectedinto rabbit to produce antiserum. Both anti Ag 3 antibody and anti Ag 7 antibody did not show any cross-reaction each other and didn't discriminate each antigen.
Another study was undertaken to characterize the difference in liver cytosol proteins between lean and obese Zucker rats. It was shown that unidentified 120 kDa protein which exist in liver cytosol fraction, but not seen in mitochondorial fraction was increased to several folds in genetically obese Zucker rats. But, this 120 kDa protein was induced by refeeding of high carbohydrate diet overnight followed after fasting for 48 hr of lean Zucker rats. One of the characterization of 120 kDa protein is similar to the receptor of apolipoprotein E reported by other investigator. These observations strongly suggested that 120 kDa protein seen in liver cytosol fraction may be responsible for lipogenesis in obese Zucker rats.
