Research Abstract |
We have applied adoptive immunotherapy by LAK cells. We maked three studies for the purpose of using effectively adoptive immunotherapy. Study 1 : LAK cells induced from PBL patierits with scqueamous carcinoma and of normal human, cytotoxic activity and lymphocyte subsets after culture were compared two kinds of LAK cells. In LAK cells from PBL of scqueamous carcinoma patients, cytotoxic activity against K562 and Daudi was as high as activity from normal human. Study 2 : We tried to augment cytotoxic activity by using PWM or PHA after induction of LAK cells. Augmentaion of cytotoxic activity against K562 was shown by using both PWM and PHA. But augmentaion of cytotoxic activity against Daudi was shown by using PHA only. Study 3 : We tried new method for assay of cytotoxic activity by using cFDA and flowcytometry. This new method showed correlation to Cr release assay.
