Research Abstract |
When human fibroblast cells were cultured in a multiplate either covered with a lid or, alternatively without a lid, proliferation of the cells in the plate without a lid was significantly less as compared with that in the plate with a lid. The explanation of the result may be as follows : A highly volatile growth factor (s) was secreted from the cells or bovine calf serum. We thus tried to isolate and identify the factor (s). Fetal bovine calf serum (FCS) was freez-dried, from which the volatile part was firstly trapped in an ethanol-trap (-40 C), and further trapped in a liquid nitrogen-trap (-195.8 C). Strongly smelling and highly volatile liquid was collected in the second trap. The liquid thus collected was then processed on a gasschromatography-massspectrometer (GC-MS). As a result 4 known molecules : ethanol, acetone, ammonia, and cyclohexanol were identified, however, several unidentified molecules seemed to be remained. Those four identified molecules possessed unfortunately no growth promoting effects on the cells. Unidentified molecules have been tried to processed on further steps such as a MS-MS.
