Research Abstract

自己免疫発症機序の詳細については未だ不明である。本研究で我々はトキソプラズマ原虫感染によって抗HSP70自己抗体産生が誘導され、この抗HSP70自己抗体がB-1細胞によって産生されることを明らかにした。さらに、トキソプラズマ感染により誘導されるTgHSP70特異的B-1細胞のシグナル活性化について解析を加えたその結巣、TLR4を介し、かつMyD88,IRAK4非依存性の経路により活性化されることが明らかになった。抗HSP70自己抗体を産生するB-1細胞とP3U1細胞との融合で得られたハイブリドーマが産生したモノクローナル抗体投与により抵抗性マウスがトキソプラズマ感染死することから、抗HSP70自己抗体産生B-1細胞が産生する自己抗休は宿主感染防御反応を抑制することが示された。さらに興味あることに、自己抗体非産生性B-1細胞をB細胞欠損マウスでトキソプラズマ感染に感受性を示すμMTマウスに移入すると抵抗性を獲得することが示された。このことから抗体非産生性B-1細胞は逆にトキソプラズマ感染に対する防御エフェクター細胞として機能することが明らかになった。
今後、トキソプラズマ感染により活性化されたTh1型生体反応と自己反応性B-1細胞の活性化機序および活性化B-1細胞の宿主防御反応における役割について解析を進める予定である。
