Research Abstract

In August 1991, five Japanese scientists had visited Malaysia and performed the project with the staff members of Universiti Pertanian Malaysia (UPM) and Malaysia Agricultural Research & Development Institute (MARDI). Under our project, four experiments were carried out. Those were in vitro fertilization in buffalo and indigenous cattle, vitrification of bovine embryos, chromosome analysis in buffalo, and pulsatile patterns of LH secretion in sheep. Results from these four experiments are summarized as follows.
1. We have demonstrated for the first time that ovarian oocytes from prepuberal swamp buffalo and Kedah-Kelantan (K-K) calves can mature and be fertilized normally in vitro using an in vitro fertilization procedure generally applied in Bos taurus. Normal fertilization for K-K calves was 50%. About 20-% of inseminated oocytes from swamp buffalo developed into the morula stage. To establish a reliable system for embryo production in the water buffalo and the zebu, further studies on IVM, IVF and culturing of fertilized embryos in vitro are in progress in UPM.
2. Embryos recovered from Sahiwal x Friesian crossbred cows were vitrified using our new vitrification solution which contains 20% ethylene glycol, 20% DMSO and 10% 1, 3-butanediol. After warming they were transferred to recipient animals, and we have obtained pregnancies. To confirm the reliability of our new vitrification procedure, further experiments are now conducted in MARDI.
3. The high-resolution GTG-banding was conducted for detailed observation of the bands in chromosome 1 in the swamp buffalo. The swamp chromosome 1 revealed the tandem fusion between chromosomes 4 and 9 in the river buffalo. The region of the fusion was indicated as band 1q19 or interband 1q19-1q110 in 400-, 500-and 650-bands stages.
4. The acute effects of the introduction of rams on the pulsatile patterns of LH secretion in postpartum anestrous sheep was examined. Collected serum samples are now being checked by RIA.
