Research Abstract

The objective of this project is to isolate and identify cell adhesion molecules which are present on the surface of hydra cells, and to examine the roles which these molecules play in morphogenesis of hydra tissue.
One can consider pattern formation process in hydra to consist of two sub-processes : prepattern formation and morphogenesis. By the former sub-process, regional specification occurs in various tissue regions by some type of information (positional information) present in the tissue. This determines the types of structures (head, foot, bud, etc.) which will be eventually formed at various regions. By the latter sub-process, various structures are actually formed based on the specification made by the former sub-process.
In previous studies we examined mainly pre-pattern formation process in hydra. In recent years, we have begun also to examine the morphogenetic process. In this project, we have cooperated with Prof. C. N. David at the Zoological Institute of University of Mft … More nchen and Prof. H. R. Bode at the Developmental Biology Center of University of California at Irvine to look for monoclonal antibodies which will bind specifically to the cell adhesion molecules present on the hydra cells, and thereby block formation of the reaggregate in the hydra cell dissociation-reaggregation system. We have obtained a number of monoclonal antibodies which specifically recognize antigens present on the surface of various cell types including ectodermal epithelial cells, endodermal epithelial cells or interstitial stem cells. The roles of these antigens in cell adhesion and morphogenesis are currently under investigation.
We have also started a project to isolate hydra homologous of vertebrate cadherin genes. A clone was isolated from hydra cDNA by PCR method using degenerate primers for consensus amino acid sequences near the C-terminal region of vertebrate eadherins. The clone isolated has about 150 bp, and its sequence shows a high degree of homology to vertebrate cadherins.
