Research Abstract

染色体分配の分子機構を明らかにするため、平賀は大腸菌のMukB蛋白質(177.7kDa)を精製し、この蛋白がDNA結合能および亜鉛存在下でのATP,GTP結合能を持つことを明らかにした。加藤は大腸菌のDNAトポイソメラ-ゼIVと内膜との結合はDNase感受性であるが、DNAジャイレ-スと内膜との結合はDNase非感受性であることを明らかにした。DNAジャイレ-スは染色体上のREP配列に特異的に結合するが、topoIVではREP配列に対する特異性は見られなかった。菊池はヒトII型DNAポイソメラ-ゼの細胞周期に伴う細胞内分布の変動を明らかにした。西村(行)は大腸菌のpar変異株における蛋白リン酸化反応を調べ、par‐530では80kDaのリン酸化蛋白を欠くことを発見した。西村(昭)は細胞分裂の頻度決定に関するcfcA変異がグリシン‐tRNA合成酵素αサブユニットの遺伝子で相補されることを明らかにした。岡崎はヒト染色体セントロメア領域に存在するアルフォイド反復配列中の17bp配列(CENP‐Bbox)に特異的に結合する80kDaの蛋白2分子がCENP‐Bboxを持つ2分子のDNAと複合体を作ることを明らかにした。丹羽は分裂酵母の染色体動原体領域のdg‐dh配列は姉妹染色体の結合制御(特に減数分裂において)に重要な役割を果たすことを明らかにした。永井は大腸菌染色体複製起点と複合体を作る17kDaの蛋白を発見した。小椋はFtsH蛋白質(70kDa)がATP結合配列を持つ膜蛋白であり、PBP3やβ‐lactamaseの膜輸送に関与することを明らかにした。公募研究の三木はFプラスミドのLetD蛋白がDNAジャイレ-ス活性(Aサブユニット活性)を阻害し、LetA蛋白がその阻害を回復させることを明らかにした。公募研究の岸本はG2期末ではcdc2・サイクリンB複合体は細胞質のみに存在するがその後サイクリンBは活性型に変換され一部核内へ移行し、核膜の崩壊が起きることを明らかにした。
