Research Abstract

本研究の目的はパ-キンソン病などの神経変性疾患を惹起する内因性、外因性の物質の発見と神経毒性の機序を解明することにある。本年度はド-パミン(DA)細胞への毒性を持つものとしてDAとアルデヒドから生成されるSalsolinol(SAL)等の6,7ーDihydroxyー1,2,3,4ーtetrahydroisoquinoline(DHTIQ)を検討した。SALをより毒性の高い物質へと生成する代謝系が脳内に存在することを明かとした。ラット脳でmicrodialysis法によりDHTIQを還流したところDHTIQがNーメチル化されることと、脳の部位では黒質に有意にNーメチル転移酵素活性が高いことを見いだした。(Maruyama et al.,J.Neurochem.印刷中)。これらNーメチル化したDHTIQはヒト脳での存在がGCーMassにより確認された(Niwa et al.,1991)。さらにDHTIQの還流によりDAとセロトニン(5ーHT)などの神経伝達物質が放出された。また部分精製した酵素サンプルでの実験でDHTIQはDAの生合成の律速酵素であるチロシン水酸化酵素(TH,Minami et al.,J.Neurochem.印刷中)を阻害した。この阻害はTHの補酵素ビオプテリンによるアロステリック効果(Minami et al.,1992)を変化させることによる。また5ーHTの律速酵素トリプトファン水酸化酵素を碁質Lートリプトファンと非競合的にまたビオプテリンにたいし競合的に阻害した(Ota et al.,Neuropharmacology印刷中)。THまた培養細胞PC12hを用い培地にDHTIQを添加しDA細胞に対する慢性の作用を検討した所、DHTIQは100μM以上の高濃度で細胞の変性とTH活性を選択的に低下させNーメチル体が細胞毒性が高かった。またPC12h細胞へのDHTIQの取り込みがDAの再取り込み機構によることが確かめられた。これらの結果はDA細胞で生成されたDHTIQが毒性の高いものにと代謝され神経毒性を発現する可能性を示している。
