Research Abstract |
(1) DEVELOPMENT OF PHENOTYPE DETERMINATION METHOD FROM DNA WE DEVELOPED A NEW SIMPLE AND RELIABLE METHOD FOR THE DETERMINATION OF O-GENE FROM DNA SAMPLES.DNA WAS EXTRACTED BY SDS-PROTEINASE K-PHENOL METHOD OR BY CHELEX-100 METHOD.O-GENE WAS AMPLIFIED BY ALLELE SPECIFIC POLYMERASE CHAIN REACTION.THE AMPLIFIED DNA WAS DETECTED BY 15% PAGE AND SILVER NITRATE STAINING.DNA FROM UNRELATED TWENTY-FIVE O PHENOTYPE PERSONS HAD O-GENE PRODUCT, AND THOSE FROM TEN AB PHENOTYPE PERSONS HAD NO PRODUCT. (2) SEROLOGIC STUDY OF ABO PHENOTYPE AND FAMILY ANALYSIS SIXTY-FIVE SAMPLES FROM NINETEEN FAMILIES AND UNRELATED THREE HUNDRED EIGHTY-FIVE SAMPLES WERE COLLECTED IN THESE THREE YEARS.THERE WAS NO AA OR BB GENOTYPE PERSON FROM OUR SEROLOGIC AND FAMILY STUDIES. (3) FACTOR(S) AFFECTING ANTIGEN EXPRESSION FROM GENOME SEQUENCE IN THE ABO SYSTEM WE HAVE NO RESULT ON THIS SUBJECT IN THIS PERIOD.WE NEED MORE STUDIES TO GET THE ANSWER.
