Research Abstract |
Periodontium is always stressed by the mastication or oclussion which might be expressed with the several categories of mechanical stresses. In this research project, we have focuesed on the cell growth and differentiation, and cell attachment and spreading factors (CASFs) of the fibroblasts derived from human periodontal ligaments of first premolars as descrived by Kawase, T et al (1988). The CASFs which may promote the regeneration of periodontium were dramatically synthesized by the mechanical stretching. The characterization of CASFs were investgated by the following experiments. 1. The CASFs in the conditioned medium of HPLF(HPLF-CM) were not inhibited by Gly-Arg-Asp-Ser peptide containing RGD cell attachment signal domain. 2. The CASFs of HPLF-CM did not affect polyclonal antibodies against fibronectin(FN), vitronectin(VN) and their receptors and could bind with hydroxyapatite. 3. The HPLF-CM did not contain only CASFs, but also inhibitors to CASF activity which were regulated by growth factors. We conclude that mechanical stress may play an important role in the cell proliferation and differentiation, and also could regulate the production of extracellular matrix such as CASFs.
