Research Abstract |
In vivo expression system using Xenopus oocyte by injecting polyA or total RNA extracted from labellar taste organs of the fleshfly has been established. An inward current was observed when the egg was perf used with sugar stimulants that stimulate fly taste receptors. In most cases the response was much higher for sugars like sucrose or glucose and smaller or absent for non-stimulating sugars like mannose or lactose. Genomic fragments that possibly cover the Drosophila taste gene Tre were isolated from YAC clones. Since the fragment contains more than 100 Kb length we are currently subcloning smaller fragments to identify the gene. The effect of Tre were analyzed physiologically by recording the labellar sensory neurons and compared with behavioral results that had been published earlier.
