前立腺がんモデルを用いた前立腺発がん機構の研究

Research Abstract

F344雄ラットに3,2'-dimethyl-4-amonobiphenyl(DMAB)を発癌物質として2週毎に1回20回投与し、60週間で終了する実験モデルを用いて下記の成果を得た。
1.DMAB投与開始時と投与終了時にあるいは終了後にカドミウムを一回ないし数回筋肉内投与したところ,ラット前立腺の腹葉の非浸潤がんの発生率および発生個数が有意に上昇し,カドミウムがDMAB前立腺発がんにおいて促進的に働くことが明かとなり,カドミウムによるヒト前立腺がんの発生において未知の発がん物質との協調的作用の可能性が示唆された。
2.DMAB投与後3つの異なった用量でtestosterone propionate(TP)をシリコンチューブにて連続投与したところ、TPの用量に依存して前立腺側葉と前葉および精嚢から浸潤がんの発生が増加した.しかし前立腺腹葉のがん(非浸潤がん)の発生は逆にTP投与によって抑制され,腹葉のがんとその他の部位のがんとは生物学的に異なることが示された.
3.DMAB投与後TPとetinylestraiol(EE)を同時に投与し,側葉と前葉の浸潤癌がきわめて強く促進されることが判明し,前立腺がん発生におけるエストロゲンの役割が明かとなった.しかしdehydortestoteroneとEEにはその作用はなくエストロゲンの促進作用には充分量のテストステロンが必要であることが示唆された.
4.腹葉の非浸潤がん6例と背側葉の浸潤がん9例についてH-rasとK-rasおよびp53について点突然変異の有無を検討したところ,K-rasのCodon12にそれぞれに1例、またp53はそれぞれ2例と1例に変異を認めた.K-rasとp53に関して両腫瘍に於ける分子生物学的差異はみとめられず,現在N-rasとneuの突然変異の有無を検討中である.
