Research Abstract

(1)オオムギの根のムギネ酸合成経路のうち、ニコチアナミン合成酵素とニコチアナミンアミノ基転移酵素が、鉄欠乏処理によって顕著に誘導されること、ムギネ酸鉄の添加により一日で活性が1/2以下になることが明らかになった。
(2)鉄欠乏条件下でオオムギの根に出現する"d"タンパク質の抗体を作成し種々のイネ科植物の鉄欠乏条件下での存在を見たところ、いずれのイネ科植物においても検出された。したがって"d"スポットはイネ科に共通の鉄欠乏特有のタンパクであることが分かった。
(3)鉄欠乏条件下でオオムギの根に特異的に発現する遺伝子Ids3(dioxygenase)について、その発現部位をアンチセンスRNAを用いたin situ hybridization法を行ったところ、発現部位は内鞘内部の外側寄りであった。一方、高エネルギー研のシンクロトロンによって根の横断面の鉄の存在部位が内鞘であることが初めて証明された。したがって、Ids3はこの部位での鉄の増減に感応して発現調節されていることが明かとなった。
Ids3を大腸菌で発現させたタンパクIDS3に対して抗体を作成し、種々のイネ科植物での鉄欠乏処理区での発現を見たところ、鉄欠乏のオオムギの根においてのみIDS3タンパクが検出された。したがって、Ids3は鉄欠乏のオオムギの根にのみ特異的な遺伝子であることが分かった。
