Research Abstract

森島らP450モデルとして前年までの近位His-Cys変異体以外に新たに近位His-Cys、遠位His-Valダブル変異体を作成し、過酸化物のO-O結合のヘテロリシス開裂におよぼす遠位ヒスチジンの効果を検討した。その結果、遠位ヒスチジンの顕著な効果は見られなかった。また、サブユニット界面のいくつかのアミノ酸の置換によるヘモグロビンの高次構造変化と配位子結合反応特性を検討した。さらに、モジュール置換のキメラ型ヘモグロビンを作成し、サブユニットの会合特性を調べた。石村らはP-450のヘム近傍のThr252の隣のAsp251を種々のアミノ酸に置換した変異体を用いて酸素添加反応活性を検討した。さらに、T252Aを用いて初めてP450CompoundIの吸収スペクトルの測定に成功した。三木らは前年度に引き続きチトクロムC2について分子動力学をとりいれた結晶構造のさらなる精密化をおこなった。阿久津らはチトクロムC3のNMRスペクトルのシグナル特にヘム鉄に配位したヒスチジンのプロトンシグナルの帰属などを行ない、四つのヘムのヘム近傍ミクロ構造と各々のヘムの酸化還元電位との関係を明らかにした。上山らは鉄硫黄蛋白質の活性中心モデルとして二重のNH…S水素結合をもつ錯体を合成しその重要性を示した。小林らはパルスラジオリシス法を駆使して蛋白質内電子移動過程を調べると共に、P-450モデルとしてのミオグロビンのHis-Cys変異体と過酸化水素との反応活性種を調べた。福住らはNADHの電子移動酸化によって生成するラジカルカチオンの反応性ならびに酸素への電子移動に対する触媒作用を詳細に調べた。折井は光励起NAD(P)HによるチトクロムCの還元反応の電子移動過程を詳細に調べた。
