Research Abstract

許容温度(35℃)では,トランフォーム能を有するが,非許容温度(39℃)では,トランスフォーム能を大きく洞弱させる温度感受性変異Abelsonウイルス(ts A-MulV)の全ゲノムのDNA sequenceから,v-ablのkinase domain内の566番目のアミノ酸のロイシンからアルギニンへの変異が,温度感受性を示す原因であることがわかった。このts v-ablをクローニングし,retouviral vectorにつないだ。このretroviral vectorのLTRは,Mo-MuLVのLTRとは異なり,未分化細胞であるEC細胞中でも発現しうるMPSVを用いた.このretrovir
al vectorを,5-FUで処理したマウスの骨髄細胞に,in vitroで感染させ(lL-6+SCFの条件下で2日間培養→感染→さらに2日間同条件で培養),致死的に放射線照射マウスに,骨髄移植を行い,約1カ月後,骨髄細胞や脾細胞を,lL-6+SCF下で,許容温度(35℃)で培養し,cell line化をはかった。このように方法で樹立されたcell line,ASC-1は,許容温度では,芽球様の形態を示し,c-Kit,Sca-1,Thy-1及び,すべてのlineage markerは,陰性であったが,非許容温度で一週間培養すると,Gr-1やSca-1が出現した。また,致死的に放射線照射したマ
ウスに,このcell lineを輸注したところ,このcell lineは,Gr-1,Mac-1,TER119,Thy-1などのマーカーをもった細胞に分化することがわかった。以上より,このASC-1は,かなり未熟な細胞株であることが示唆された。
