Research Abstract

心筋のイオンチャンネルの開閉・調節機序を、電気生理学的および分子生物学的に研究し、以下に報告する多彩な成果が挙げられた。(1)Caチャンネル:ヒトの心房筋のL型Ca電流は5-HTによりチャンネル利用率増大によって増強された。心臓由来株化培養細胞H9c2では心筋型と骨格筋型のL型Caチャンネルが発現していた。心筋Caチャンネルの活性維持にはカルパスタチンのN-末端が重要な役割をする。カネパスタチンの効果は、ATP、A-キナーゼ、その他の細胞内物質によって増強され、複数の細胞内因子の関与が判明した。カエル心室筋においてCa依存性のCa電流増強現象には、リン酸化とは独立に細胞内Mgが深く関わっていた。(2)Naチャンネル:カエル心室筋で種々のリン酸化を試みたが、リン酸化過程による制御はなかった。(3)Clチャンネル:βアドレナリン受容体刺激で活性化されるCl電流をムスカリン受容体刺激はアデニルシクラーゼ(AC)活性抑制によって減少した。またcGMPの細胞内投与はPDE阻害によってCl電流を増大した。(4)血管平滑筋Caチャンネル:ラット冠動脈平滑筋のCa電流はα-およびβ-受容体刺激により増強された。この増強はG蛋白、または未知のセカンドメッセンジャーを介してもたらされる、ACは関与せずCキナーゼは補完的に働いていると考えられた。(5)伸展受容(SA)チャンネル:培養血管内皮細胞において、単一チャンネル電流と全細胞電流でSAチャンネルの活性化に必要な膜張力は約10dyne/cmと一致した。従ってSAチャンネルは機械刺激に対する細胞応答に直接関わる。(6)Kチャンネル:ラット心筋よりクローニングした2種類のKチャンネル、Kv1.2、Kv1.4、はPI代謝回転の亢進を起こす受容体刺激により抑制され、その抑制の一部にはCキナーゼの活性化が関与している。
