Research Abstract

細胞の増殖・癌化また細胞死に重要な役割を果たす細胞内酸化還元状態(細胞内レッドクス)は細胞内で産生される種々の活性酸素のみならず細胞外からの酸化的ストレスによっても強い影響を受ける。我々がHTLV-1感染細胞株培養上清よりIL-2 receptor α鎖誘導因子として分離精製した多機能蛋白ATL由来因子(ADF)は大腸菌の還元補酵素thioredoxin(TRX)のヒトhomologueであり、その活性中心に存在する二つのCys-SH残基を介した強い還元活性を示し、glutathione系と共に細胞内の還元状態を司る抗酸化ストレス因子である。これまでにADF/TRXの細胞内での作用としては細胞内蛋白に対するPDI活性、活性酸素除去に必要とされるcatalase様活性を持つことが我々を含めた幾つかの研究グループにより明かとなっている。本研究ではADF/TRXの還元活性に注目し、ADF/TRXを含めた細胞内酸化還元調節因子と細胞内シグナル伝達機構との関連について、特に転写制御因子及び細胞内蛋白のチロシン燐酸化を中心に検討を行うことを目的とした。その結果ADF/TRXを含む還元因子が、転写因子NF-kBのDNAへの結合の増強を、また酸化状態が細胞内チロシンリン酸化を誘導することが判明した。本研究により細胞内外のredox状態が細胞内シグナル伝達に深く関与していることが明らかとなった。
