Research Abstract

1.酵母の酸性化コンパートメントの形態構築に関わるVAM4,VAM2,VAM6の3遺伝子の構造を明らかにした.VAM4遺伝子の機能改変の結果,GTP-GDPサイクルがVAM4の機能に重要であることを示した.さらに,酵母の液胞形成に必須なVam3Pのシロイヌナズナ相同遺伝子を同定,単離した.
2.V-ATPaseの選択的阻害剤Folimycinは,小胞体からは出ていくがゴルジ器官シス槽での糖鎖プロセシングを受ける以前の過程で糖蛋白質の細胞内転送を阻害すること,また,本阻害剤はV0ドメインに結合することによって阻害作用を示すことを明らかにした.更に,既知のV-ATPase阻害剤と異なって非常に可逆性に富むDestruxin Bの新規な阻害作用を見い出した.V-ATPaseの細胞機能を解析するうえで大変有用となる発見である.
3.V-ATPaseによる貧食胞の酸性化が,網膜色素上皮細胞による視細胞外節の分解に必須であることがV-ATPase阻害剤を用いて示された.
4.マウス液胞型ATPaseプロテオリピッドの遺伝子をクローン化しエクソン/イントロン構造を明らかにした.またあわせて2つの偽遺伝子の構造が決定された.これらの結果をもとにターゲッティングベクターを作成し,ES細胞に導入した.トランスフォーメーションにより,リソソーム内pHの上昇が誘起されることを見い出した.これらの一連の研究結果により,細胞内酸性化コンパートメントの形成においても,神経細胞から酵母に至るまでの多様な細胞で小胞輸送に関与している細胞構築に関わる分子装置のサブセットが機能していることが示された.
5.連鎖球菌Na^+輸送性V-ATPaseがNa^+のuniportを行う酵素であること,16kDaプロテオリピッド(NtpK)に存在するDCCD結合性Glu-139がNa^+の認識に重要な役割を果たすことを明らかにした.本酵素オペロンの発現系を構築した.
6.好塩性古細菌のV-ATPaseの分子量30KDa(γ),20KDa(δ),12KDa(ε)の3種類のポリペプチドが単離された.新たに見い出したatpF遺伝子は連鎖球菌のNa^+-ATPaseのntp Cと21.2%の相同性を示した.
