Research Abstract

家族性アルツハイマー病の遺伝子座(FAD)の解析に関しては、わが国においても第14染色体(14q24.3)に連鎖する早発型家族性アルツハイマー病家系の存在が確認され,さらにFAD遺伝子の候補領域がD14S277とD14S53の間の8cMに存在することが明らかにされた。今後その原因遺伝子の究明に大いに寄与するものと期待される。また,14q24.3領域のFAD遺伝子のポジショナルクローニングに向けて,ゲノムDNAの物理地図の作製と,同領域における発現遺伝子の解析が順調に進んでおり,3つの新たな候補遺伝子の同定がなされ,現在FAD遺伝子かどうかの解析が行われている。
アルツハイマー病と同様の脳病理所見を示すダウン症については,第21染色体のダウン症関連領域(DS領域)の1.8Mb領域について約94%をカバーする最大約900kbの4つのコンティーグを完成させた。さらに同領域から59個のエクソン,14種のcDNAを単離した。
アルツハイマー病の危険因子としてのApoE4に関しては,従来から言われていた遅発型アルツハイマー病の危険因子であるばかりでなく,早発型アルツハイマー病を含めてアルツハイマー病に共通する危険因子であることを証明した。またApoE4の存在が側頭葉病変を強くすること,老人脳においてアルツハイマー病的な変化が加速されて出現することを見出した。
アミロイドβタンパクについては,Aβ(1-40)Aβ(1-42)の発現系を用いて検討したところ,Aβ(1-40)は細胞外に分泌されやすいのに対して,Aβ(1-42)は細胞内に蓄積しやすいことが判明し、アルツハイマー病におけるAβ(1-42)の重要性が示唆された。
