Research Abstract

本研究は、主としてアルツハイマー病を中心的課題として取り上げ、老年痴呆の分子機構にアプローチしようとするものである。全三班の中で、本研究班は主要研究目的として神経栄養因子を取り上げ、この蛋白質群の神経細胞の生存と機能維持の細胞生物学的意義を明らかにすることから、神経細胞変性と死のメカニズムを解明しようと考えた。その結果、(1)アルツハイマー病との関連が予想されている脳由来神経栄養因子(BDNF)など、ニューロトロフィンによる神経細胞の生存について生後ラッド中枢神経細胞の培養系を用いる解析がすすんだ。
(2)ニューロトロフィン-3(NT-3)の高感度酵素免疫測定法の開発を終え、NT-3は海馬のCA1領域にはほとんどなく、存在の局在が明らかになった。また、BDNFについての測定法も完成に近づいている。
(3)ミクログリア細胞など、各種グリア細胞の選択的培養から、新しい神経栄養因子活性をもつ蛋白質の精製に成功した。
(4)成長抑制因子(GIF)の新しい活性を明らかにした。また、アポプロテインEの作用との関連を明らかにした。
(5)神経突起伸展因子レセプターのクローニングに成功した。
