Research Abstract

長期増強(LTP)現象は、記憶形成の基礎過程であるシナプスの可塑性の一つとして注目されているが、我々は視覚-運動系の統合調節に重要な役割を果たしている上丘において、顕著なLTPが惹起されることを世界で初めて報告した(Okada,1990)。本研究課題はこのLTP発現の分子機構と生理的意義の解明であったが、2年間にわたる研究成果として、
1.上丘LTP発現にはグルタミン酸受容体のNMDA受容体が関与しているが、その誘発初期にはカルモジュリン(CAM)系が、維持過程には蛋白キナーゼC(PKC)系が関与していることが明らかとなった。
2.マイクロダイアリシスを用いて上丘のグルタミン酸放出を測定する研究から、上丘LTPの発現時にシナプス前性に放出される興奮性伝達物質グルタミン酸量が増大していることが明らかとなった。
3.上丘浅灰白質層単離培養ニューロンのパッチクランプ法による解析から、このニューロンはグルタミン酸感受性であることが明らかとなった。
4.アデノシンおよびその誘導体を上丘浅灰白質層に作用させるとLTPと同様の現象が惹起されるが、この時グルタミン酸の放出が増大していること、その維持にPKC系およびPKA系が関与しており、LTPとアデノシンによるLTP作用の類似性が明らかとなった。
5.同側の大脳皮質視覚野から上丘への投射が上丘ニューロンを活性化し、それがGABA作動性ニューロンを介して視神経からのLTP形成を抑制していることが明らかとなり、上丘の神経回路網におけるLTPの生理的意義の解明に重要な糸口がつかめた。
