Research Abstract

最近、mutator phenotypeないしgenomic instabilityが多段階発癌の進行モーメントとして重要視されている。B型肝炎ウイルス(HBV)は宿主の染色体転座(Hino,O.et al.Proc.Natl.Acad.Sci.USA 83.8338‐8342,1986)を誘発し、さらにHBVのpre‐core領域には、増殖期の細胞抽出蛋白存在下でDNA組換え(in vitro)を促進する働きがある(Hino,O.,et al.Proc.Natl.Acad.Sci.USA 88:9248‐9252,1991)。そこで我々は、HBVによる肝発癌過程において、同ウイルスが積極的に染色体不安定性を亢進させているとの作業仮説を立て研究を進めている。今回、我々は、増殖期細胞(マウス白血病細胞70/Z3)抽出蛋白存在下でrecombination 活性があり、なおかつ同蛋白との特異的結合能があるHBVのpre‐core領域内の sequenceを決定した(Kajino,K.and Hino,O.submitted)。現在、そのsequenceを用いてSouth‐Westerm法により結合蛋白の単離、同定を行っている。
