Research Abstract |
Composite materials with Hydroxyapatite and Bone Morphogenetic Protein extracted from bovine bone were implanted into adult dogs using vessel attached incubating chambers. Histological examinations of ectopic bone formations in the implanted materials were carried out. The summary was as follows ; 1. In the BMP-HAP group, immature, newly-induced bones were found adjacent to the HAP particles after 28 days. These bones increased in volume gradually, but numerous lymphocytes and histiocytes were found in the interparticle tissues throughout the observed term. Proliferation of granulation tissues consisting of fibroblasts and capillaries seemed to be lower in this group compared to the other groups. 2. In the HAP only group, newly formed bone was found in the focal interstitial tissues in the specimens after 150 days. This finding suggested that HAP has not only osteoconductive abilities but also osteoinductive abilities. 3. In the HAP-autogenous bone chips group, 28 days after implantation

newly induced bone could be seen bordering the implanted bone. The induced bone infiltrated into the HAP particle-tissue interface. Bone marrow formation was found in the specimens after 60 days. 4. The vessel-attached chamber which we designed was a useful means for examining the ectopic bone formation. Under these conditions, it was clarified that induced bone formation associated with BMP or autogenous bone chips showed not only cartilage ossification but also fibrous ossification ; the mechanism of ossification depends on the circumstances or carrier of bone matrix. In the future, it is important to investigate the circumstances of bone formation site and carrier materials, and osteogenic cell differentiation of the host in order to clarify the origin of the aggregated undifferentiated mesenchymal cells. Our results indicated that it is important to develop a new type of carrier which will be able to compound with BMP and be used as artificial bone materials to treat bone defects effectively. Less
