Research Abstract

心臓血管中枢の機能を調節する内因性物質として、強力な内因性ペプチドであるエンドセリン(ET)と内因性物質であるが経口摂取により補充されるカルシウムイオン(Ca^<2+>)をとりあげて研究した。
ETに関しては、Kuriharaたちがgene targetingの方法で作成したET-1遺伝子の欠損マウスにおいて、心臓血管中枢・呼吸中枢の性質を検討した。ET-1欠損マウスのホモ接合体は、出生時に呼吸開始が不能のため死亡し、また第一鰓弓由来の器官に奇形が見られた。他方、ヘテロ接合体は、血中および組織中のET-1濃度が同腹の野性型マウスに比べて約1/2であるが、正常に成長・繁殖をした。しかしヘテロ接合体の血圧は、野性型に比べて有意に高かった。さらに、低O_2または高CO_2負荷にたいする呼吸反射を調べて呼吸中枢の機能を検討したところ、ヘテロ接合体では呼吸中枢の機能が低下していた。また、呼吸中枢から心臓血管中枢に対する影響を、血圧のコヒーレンスを調べて検討したところ、影響の増強が見られ、心臓血管中枢の出力である血管運動神経の活動が上昇していることが示唆された。
Ca^<2+>の心臓血管中枢への影響に関しては、麻酔したラットいおいて、大動脈神経の電気刺激に対する血圧および腎交感神経活動の反射性調節を幼若の高血圧自然発症ラット(SHR)を用いて調べた。幼若SHRに高食塩負荷を行うと高血圧の発生が促進され、大動脈神経-血圧または腎交感神経活動反射の劣化がみられた。しかし、同時にCa^+を負荷すると反射の劣化は起こらなかった。つまり、Ca^<2+>は、食塩負荷による心臓血管中枢の機能劣化を防ぐ作用があることがわかった。
