Research Abstract |
In this study the characteristics and function of apoprotein A1 in cholesterol gallstone were investigated, and the following results were obtained ; (1) Biochemical characteristics of apoprotein A in cholesterol gallstone was analyzed by using SDS- PAGE and Western blot methods. Characteristics of apoprotein A in cholesterol gallstone were similar to those in hepati bile. Meanwhile, biliary aporotein was quantitatively determined by using ELISA, which was established in our laboratory. The content of apoprotein A in cholesterol gallstone was very low. Thus, biliary apoprotein A could be mobililized for preventation of cholesterol nuleation without precipitation into gallstone. (2) Measurement of apoprotein A in hepatic bile was carried out by using ELISA.There was significant correlation between bilary apoprotein A1 concentration and biliary cholesterol level. According to these data, it was demonstrated that hepatic lithogrnic bile might induce apoprotein A1 secretion into bile. (3) Study on function of apoprotein A in cholesterol gallston. Under investigation. (4) Apoprotein A1 in cholesterol gallstone was detected by histoimmunostaining, and it was shown that in cholesterol gallstone apoprotein A1 and A2 exists between cholesterol layr. These results gave us many important informations on the mechanism of cholesterol gallstone formation.
