Effect of hepatic copper on signal intensity in magnetic resonance imaging of the liver : We analyzed the signal intensity of the liver of LEC rats, which have an abnormal copper accumulation in the liver, and of Wistar rats, and studied the correlation between signal intensity of the liver and hepatic copper content. The signal intensity of the liver of LEC rat dose not differ from that of Wistar rats on both T1 and T2 weighted images. We concluded that copper accumulation may not be responsible for the high intensity of HCCs on T1 weighted images. Effect of copper and copper binding protein on CT attenuation value of the liver : We analyzed the effect of copper and copper binding protein on CT attenuation value in a rat model. Five Long-Evan's-cinnamon(LEC)rats, which are thought to be rat form of Wilson's disease, and 28 Wistar rats were included in this study. Wistar rats were divided into 4 groups. tional copper(cupric chloride solution, daily dose of 1.8-2.4mg copper/kg)was intraperitoneally overloaded in 7 during 32-60 days. High dose copper(cupric nitrilotriacetate, daily dose of 4 mg copper/kg)was given intraperitoneally in 11 rats during 21-120 days. Hepatic copper and iron were measured by atomic absorption spectrophotometry. CT attenuation values at non-enhanced CT of the liver were examined in all rats and compare with hepatic iron and copper content, and grade of orcein positive granules that is thought to be a counter part of polymerized methalothionein in lysosome. The difference hepatic copper and attenuation value at CT was not statistically significant(correlation coefficient=0.056, p=0.96). The difference between hepatic iron content and attenuation value was statistically significant(R=0.561, p=0.007). However, CT value has a strong positive correlation with the grade of orcein positive granules(Spearman's correlation coefficient Rho=0.755, Rho corrected for ties, 0.738, p=0.0001). CT value of the liver of LEC rats did not differ from that of normal control rats. We concluded that CT attenuation value of the liver can be elevated by rich polymerized methalothionein(copper binding protein)in lysosome and has a negative correlation with hepatic copper content.
