Research Abstract |
It is very interesting problem awaiting solution to control artificially autoimmune diseases and rejection in allo-grafting, of which detail mechanisms have been still unclear. For purposes of development of immunosuppressants with new mechanisms, extracts obtainined from thermophiles have been applied on screening test using mouse mixed lymphocyte reaction (MLR). As the results of the screening, an active fraction, Fr. 5-B, was isolated from Bacillus stearothermophilus UK563. Fr. 5-B inhibited the induction of cytotoxic T cells and expression of MHC class II antigen on mecrophages, but did not affect mitogen responses of lymphocytes. This fraction consisted mainly of 1, 3-diacylglcerols with iso- and ante-isosaturated fatty acids (n=15-17). 1, 3-Di-14-methylpentadecanoylglycerol synthesized as a putative major component of Fr. 5-B, suppressed MLR and auto MLR, and increased the inducction of suppressor T cells. Since few substances effective in controlling suppressor T cells have been reported, further studies on action mechanism of Fr. 5-B will be expected.
