Until now, K1 and K10 keratin peprides which are the differentiated marker keratins has been said not to express in cultured human squamous cell carcinoma(SCC). When keratin was extracted from SCC, the cells were treated with a high salt solution, so far. As this solution has originally emploved to analyze keratin of tumor tissues, the solution may be a little drastic against cultured cells. Therefore, we used and analyzed using a more mild and aqueous solutions for 2 kind of SCC cell lines. Surprisingly, evidence of the K1 peptide was indicated by biochemical and immunological methods, and mRNA level. To confirm this fact which is a general, whether or not, we further carried out using the other 4 kind of SCC.We obtained the above same result which is present K1 peptide in SCCs employed. Furthermore, K10 peptide which is a pair keratin of K1 peptide, was not expressed in all cell lines employed. The K1 peptide expressed was solubilyzed about 90 % in 1-2 M urea contained 5 % beta-ME. From these results, K1 peptide is present alone in SCC and many investigators may be overlooked this evidence to employ a high salt solution. For this K1 peptide may have abnormal characters from the above results, it suggested that K10 peptide was not induced in SCC cell lines.
