Research Abstract

Recent studies on hypertension are focused on the linkage analysis between blood pressure and genetic markers in hypertensive rats as well as human. In Lyon Hypertensive rat REN locus in chromosome 13 are reported to link to basal blood pressure. In SHRSP NGFR locus on chromosome 1 are shown to link to basal blood pressure. SA locus near LSN locus was also reported to relate to basal blood pressure in SHR. Salt sensitive hypertension has been shown to associate with ACE locus on chromosome 10 in SHRSP and DS rats. SA, REN, and ANF receptor loci are also reported to link to hypertension in DS rats. However, little is known about genetic locus of human hypertension. AGT, but not ACE, locus has been reported to link to blood pressure in human.
In this study we investigated the population in Port Aregre in Brazil for linkage analysis of large hypertensive families. We chose the population, since 1) there are many patients suffering hypertension, 2) they have large families being suitable for the linkage analysis, and 3) antihypertensive drugs are not so available as in Japan. Twenty-five families, 174 persons (58 men and 116 women) were examined with questionnaire. Hundred and six persons showed high blood pressure including borderline hypertension. Blood samples were obtained from 130 persons to subject to the linkage analysis based on the micro satellite polymorphism. As far as investigated, we failed to find the linkage between ACE locus and blood pressure, consisting with the results reported by other investigators.
