Research Abstract

本研究では、糖鎖遺伝子であるGnT-III遺伝子の細胞導入によりGnT-IIIの産物であるbisecting GlcNAcが糖タンパク質に付加されることにより、細胞膜に存在する糖タンパク質の選別輸送が抑制され、選別輸送が反対方向へおこり、ゴルジやリソソームに蓄積された。しかし、リソソームに存在する糖タンパク質や、分泌性の糖タンパク質の選別輸送は障害されなかった。また、PC12細胞では、神経成長因子NGFのレセプターであるTrkAの糖鎖が改変され、二量体形成やクラスタリングが抑制され、細胞核への増殖シグナルが抑制され、分化が抑制された。がんの転移に密接な関係があり、転移がん細胞で著しく発現するGnT-V遺伝子の上流解析をおこない、プロトオンコジーンの転写因子Ets-1がプロモーター活性をもつことをあきらかにした。また、新しいシアル酸転移酵素遺伝子のクローニングに成功した。新しいα1-6 Fucosyl transferaseをヒトおよびブタ組織から分離精製に成功し、その遺伝子クローニングに成功した。ルイスXおよびシアリルルイスXの生合成機構と人類遺伝学的な分布をあきらかにした。また、細胞間の接着に重要なヘパリン結合性成長因子であるミドカインのヘパラン結合の分子機構と細胞生物学的意義を明らかにした。
このように本研究では、糖鎖遺伝子を細胞に導入することにより、糖鎖遺伝子を発現させ、その産物である糖転移酵素を過剰発現させることにより、細胞内や細胞表面の糖タンパク質や糖脂質の糖鎖を改変させ、それらの新たに生成した糖鎖のもつ意義を明らかにすることに成功した。また、これらの糖鎖遺伝子の制御機構、細胞生物学的な意義、とくに細胞増殖、細胞接着、細胞間認識機構における糖鎖遺伝子のかかわりを解明した。
