Research Abstract

器官の形成と再生を制御する分子機構を明らかにするため、以下の研究を行い成果を得た。
1、マウス神経細胞接着分プレキシンファミリーに属する新たな分子プレキシン4を同定した。また、神経細胞接着分子ニューロピリンの細胞接着活性ドメインを決定した。(藤澤)
2、ニワトリ胚神経系で、en,PAX5,Wnt-1,Elf-1,RAGS相互の発現調節のカスケードを明らかにし、網膜視蓋投射パターンの形成との相関を解析した。(仲村)
3、ニワトリ受容体型チロシンキナーゼCek8とそのリガンドElf-1,RAGSが運動ニューロン軸索伸長の制御に関わっていることを明かにした(田中)
4、肢芽の間充織細胞が部位特異的な細胞接着能を持ち、この細胞接着に関与する可能性があるプロトカドヘリン2を見いだした。(井出)
5、ドミナントネガテイブBMPレセプターをニワトリ胚肢芽に導入する方法で、BMPが肢芽のプログラム細胞死を引き起こす因子であることを明らかにした。(黒岩)
6、肝細胞増殖因子HGFが肺の形態形成を制御する因子であるとの結論を得た。更に、アフリカツメガエルでHGFファミリー分子ならびにそのレセプターを同定した。(中村)
7、β-1インテグリンがコラーゲンやフィブロネクチンの受容体として、色素上皮細胞の分化状態や分化転換に重要な役割をはたすことを示した。(江口)
以上のように、本年度の研究により、神経系や四肢など高次な形態を備えた器官の形成・再生に関与する新たな分子遺伝子が見いだされ、更にこれら分子・遺伝子の機能的連関が明らかにされ、多細胞高次形態の形成の分子機構の解明が進展した。
