Research Abstract |
Two types of cDNAs encoding mouse immauoglobulin heavy (VH) and light (Vc) chain variable (V) domains against UV-induced DNA damage have been prepared. The cDNA was cloned for seyuencing and expression, and the nucleoticle sequences of the V domains were determined by PCR.Genes for a linker (Gly_4-Ser)_3 was inserted between the two domains, and the gene was expressed using the T7 promoter. The single chain Fv for pyrimidine (6-4) photo product was purified and tested for its ability to oligonucleotides containing the (6-4) photoproduct. The scFv was found to be specific.
