生物時計の光同調機序:松果体メラトニンを指標としたin vivo解析

Research Abstract

本研究は、光による直接抑制と生物時計を介する位相反応の二重の調節を受ける松果体メラトニンを指標に、ラットを用い、光同調機序を明らかにすることを目的とし、微量測定の困難なメラトニン変動を同一個体で測定するため、in vivoマイクロダイアリシス法を松果体に適応して行われた。松果体および血漿メラトニンはRIA法にて測定した。
1.メラトニン光抑制の波長と位相依存性
暗期中点と暗期後半に同一光子量の赤と緑の単色光を照射したところ、暗期中点には赤色光による抑制は殆どみられなかったが、暗期後半では両単色光とも速やかにメラトニン合成を抑制した。メラトニン合成抑制は光の波長とリズム位相に依存し、緑色光は赤色光より、暗期後半は前半より、強く抑制することが分かった。網膜ロドプシンの光吸収スペクトルの極大な緑色域にあるため、メラトニン光抑制はロドプシンを介する可能性が示唆される。
2.松果体in vivoマイクロダイアリシスによる光の二重効果の検討
松果体マイクロダイアリシス法により連続4日間の透析を行い、30分毎メラトニンを測定し、光の二重効果を検討した。その結果、同一ラットで上述の光抑制の時刻差が確かめられ、さらに光によるメラトニンリズム位相の変位が、位相前進、後退変位とも、照射の翌日には完了していることが明らかとなった。行動、体温など他の連続測定可能な表現型リズムの位相反応には約一週間の移行期があるため、松果体メラトニンは生物時計のより直接的な表現型リズムであることが分かった。
3.網膜視床下部路の伝達物質の検討
in vivoへの投与で唯一光類似作用を生じるとされるコリン作動薬カルバコールをSCN内に投与し、松果体マイクロダイアリシス法にてメラトニンリズムへの影響を検討したが、メラトニン合成抑制もリズム位相変位もみられなかった。アセチルコリンが網膜視床下部路の伝達物質である可能性は低いと考えられる。
