Research Abstract |
A novel post-transcriptional process called trans-splicing has been found in trypanosomes and nematodes. In this process an RNA sequence, called the spliced leader (SL) , is spliced into a precursor mRNA to create the 5' segment of the mature mRNA.Since both trans-splicing and cis-splicing occur in nematodes, nematodes is a good model for investigations of mechanism between cis-and trans-splicing. The present investigation was conducted to seek immature mRNAs possessing some introns with or without the SL so that the temporal relationship between cis-and trans-splicing could be examined. cDNA libraries of adult 0.volvulus were screened with two probes, the 5'untranslated region and the coding region region. The two cDNAs clones having coding region, one of five introns, and poly-A tail without SL which were isolated in the present study indicate that polyadenylation can occur before trans-splicing and before cis-splicing is complete. Cis-splicing can also occur before trans-splicing.
