Research Abstract |
In inflammatory bowel diseases, colonic epithelial cells have both class I and class II of HLA on the surface. Recent studies on HLA showed that antigens are located inside HLA as 9-13 aminoacid of peptides. Therefore, we tried to analyze those peptide inside LHA of colonic epithelial cells in patients with inflammatory bowel disease and to elucidate the nature of the antigen recognized by cytotoxic T cell (analysis of class I HLA) or represented by colonic epithelial cells (analysis of class II HLA). We obtained colonic epthelial cells from surgical specimens of patients with inflammatory bowel disease. HLA was extracted from those homogenate cells with affinity chromatography using anti-HLA antibody. We obtained the extract containing HLA.The peptides inside LHA were separated and purified by HPLC.However, its quantity and purity were not enough to analyze the structure and nature of the peptides. We continue to improve the various conditions of the process.
