Research Abstract |
beta-AMINOPROPIONITRILE (betaAPN), A LATHYROGEN THAT INHIBITS LYSYL OXIDASE ACTIVITY,WAS REPEATEDLY ADMINISTERED TO MALE RAT PUPS FROM 2 DAYS OF AGE UNTIL SACRIFICE AT FOUR WEEKS OF AGE.BY SCANNING ELECTRON MICROSCOPY,THE LUNGS OF THE TREATED RATS SHOWED A CORROSION CAST OF ALVEOLAR CAPILLARIES THAT APPEARED ABNORMAL IN THICKNESS AND CONFIGURATION ; TRANSMISSION ELECTRON MICROSCOPY SHOWED EXTENSIVE MORPHOLOGICAL CHANGES IN EXTRACELLULAR MATRIC (ECM), INCLUDING COLLAGEN, ELASTIN,GLYCOSAMINOGLYCAN AND INTERSTITIAL CELLS.QUANTITATIVE ANALYSIS REVEALED AN INCREASE IN GLYCOSAMINOGLYCAN OF UP TO 24.4%, A DECREASE IN COLLAGEN (44.5%), AND THE DISAPPEARANCE OF LIPID-LADEN INTERSTITIAL CELLS : HOWEVER,THE CHANGE IN ELASTIN WAS LIMITED TO MORPHOLOGICAL APPEARANCE.FROM THESE OBSERVATIONS,WE CONCLUDE THAT ALTERATIONS IN CAPILLARY CONFIGURATION ARE CONNECTED TO EXTENSIVE CHANGES IN THE ECM,AND THESE CHANGES IN ECM COMPONENTS MIGHT ALSO BE INVOLVED IN THE EMPHYSEMA-LIKE LUNGS INDUCED BY betaAPN.
