Research Abstract |
Methods Transient forebrain ischemia was produced in Mongolian gerbils (1), and Wistar rats(2), which were respectively divided into normthermic and hypothermic groups. Delayd neuronal death was evaluated in the hippocampal region. Stress protein and c-Fos/c-Jun products were analyzed by immunohistochemistry and Western blotting. Carbon-fourteen leucine autoradiography was also performed (1). Extracellular concentrations of dopamine (DA), adenosine (ADO) and their metabolites in the striatum were measuredusing in vivo microdialysis (2). Cisternal irrigation of cold artificial cerebrospinal fluid (ACSF) were performed in the cat. Cerebral temperature, cerebral blood flow (CBF), blood gas analysis (BGA) and systemic blood pressure (SBP) were measured (3). Results (1) Hypothermia reducedstress response after ischekmia and produced early recovery of protein synthesis, which can enhance immediate early gene expression. Increasing c-Jun synthesis could be one of the protective mechanisms and/or survival markers effected by mild hypothermia against delayd neuronal death. (2) Intraischemic hypothermia reduced the release of DA and inhibited the uptake of ADO.Postischemic hypothermia reducedthe metabolism and elevated the concentration of ADO.(3) Cold ACSF irrigation provides 2-3゚C of focal hypothermia but had no effect on SBP or BGA.Transient decrease of CBF was observedin few cases. Conclusion Focal hypothermia provided by cisternak irrigation of cold ACSF could be a beneficial treatment for cerebralischemia, especially in combination with supportive modalities mentioned above.
