Research Abstract

The knowledge as to the molecular biological mechanism of carcinogenesis is now increasing in many human cancers. In the present research, the following results were obtained.
1) Genomic analysis of CD44 variants in human urogenital carcinoma
Total RNA extracted from surgical specimen was amplified by RT-PCR method. The amplified cDNA was analized for the variants by Southern blotting. CD44 variants were recognized in 6 out of 8 transitional cell carcinoma and all of 5 renal cell carcinoma.
2) Immunohistochemocal study of CD44 variants in human urogenital carcinoma
Normal tissues and carcinoma tissues were imuunostained with monoclonal antibody of CD44 variants (V5, V6, V7, V10). In transitional cell carcinoma, CD44 variants, especially V6, were much expressed in the area near by basement membrane.
3) CD44 variants in the cells collected from urine
Southern blot analysis and immunohistochemical study were carried out with the same methods described above in the cells collected from urine. CD44 variants were also recognized in the cells by the both way.
