Research Abstract |
1 To simplifies freezing procedure of human sperm, the floating platform was developed. Sperm-KS-II cryomedium mixture was frozen at-30゚C/min, and stored in liquid nitrogen container. 2 Hipper osmotic pressure by glycerin, which involved in KS-II cryomedium, decreased sperm motility, pentoxifilline, a phosphodiesterase inhibitor, protected sperm viability. 3 To recover motile sperm, the contents in bladder was replaced with culture medium prior to retrograde ejaculation. Contamination of urine in the culture medium denatured sperm. Whereas addition of pentoxifilline in the culture medium suppressed the toxicity of urine, improving sperm motility and velocity. 4 Cryo-accumulation of human sperm provide sufficient number of motile sperm for artificial insemination. Its clinical application to infertile couples who suffered from ejaculatory disturbance resulted two successful prehnancies following artificial insemination.
