Research Abstract

1. TIMP concentration of maternal serum didn't show any significant changes dur-ing pregnancy. It either didn't show any significant differences to non-pregnant serum. In the cases of PROM and premature labor, there were not any difference in serum levels of TIMP.The level of TIMP at delivery in amni-otic fluid was significanty (p<0.01) higher than that of maternal serum, um-bilical serum. TIMP was clearly localized in fetal membrane, amniotic epithelium, amniotic fibroblast, chorionic epithelium and de-cidua, by immunohistochemical method.
2. MMP-1 concentration of maternal serum was increased at the 3rd trimester.especially at the start of cervical maturation. The level of MMP-1 at delivery in amniotic fluid was same level to that of maternal serum, um-bilical serum..MMP-1 was clearly localized, like TIMP,in fetal membrane, amniotic epithelium, amniotic fibroblast, chorionic epithelium and decidua, by immunohistochemical method.
3. The effects of IL-1, LI-2, IL-6, TNF on the production of matrix metalloproteinases-1, TIMP-1 in cultured human amniotic and chorionic cells, were examined.In chorionic cells, IL-1, IL-2, IL-6 enhanced the production of MMP-1 and TIMP-1. TNF,also, enhanced the production of MMP-1, but supressed TIMP-1 production.
In amniotic cells, IL-1, IL-2, IL-6, TNF enhanced the production of TIMP-1, but MMP-1 was not found.
